An Experimental Study on Catecholamine Levels in the Uterus and Liver of Mature Female Rabbits by Chlorpromazine Administration


Abstract
Chlorpromazine, one of the antihistamine drugs, has been used for hypertensive disorders of pregnancy, menopausal syndrome, dysmenorrhea, and pre-menstrual tension. The drug is able to induce a number of untoward side effects, such as deleterious side effects to the hypothalamus-
pituitary-gonadal-uterine axis, pituitary-thyrotropic axis, and growth hormone.
An attempt was made to clarify the effects of chlorpromazine on the catecholamine levels in the uterus and liver of mature female albino rabbits weighing around 2kg in body weight.
A group of albino rabbits conditioned with 50 I. U. /kg of HCG for 5 days and another group with 25 mg/kg of chlorpromazine plus 50 I. U. /kg of HCG for 5 days were observed in 24 hours after conditioning. Catecholamine levels were determined by Shore and Olin method.
The results obtained were as follows,
1. The catecholamine level in the normal tissue was 0.400.12 ug/gm, while that of liver tissue were 0.170.03 fig/gm. Therefore, the catecholamine level in the uterine tissue was much higher than that in the liver tissue.
2. The catecholamine level in the Uterine tissue of the HCG conditioned group showed 0.130.05 pg/gm, indicating a decrease in its level as compared to the normal group, whereas its level in the liver showed 0.15}0.02 pg/gm, indicating no significant change.
3. The catecholamine level in the uterine tissue of the chlorpromazine plus HCG conditioned group was 0.150.03 fig/gm, indicating a decrease and its level in the liver 0.100.02 jug/gm, showing a significant decrease.
4. The catecholamine levels in the uterine ;tissue of both the HCG cooditionedlgroup and the chlorpromazine plus HCG conditioned group showed a marked decrease.
5. The_ catecholamine level in the liver tissue of the HCG conditioned group showed no significant change, on the other hand, a slight decrease.in.its level of the chlorpromazine plus HCG conditioned group was noted.
